Abstract

“Immunosenescence” is an imprecise term used to describe deleterious age-associated changes to immune parameters observed in all mammals studied so far. It represents a rapidly progressing science in the aging field, with a vertiginous volume of new data, knowledge and concepts concerning these changes. Indeed, during the almost 10 years that have passed since the first edition of this Handbook, a wealth of additional information has emerged from animal models and also from increasingly available human data. Numerous new aspects of the immune changes with aging have emerged, while others became less prominent. The application of the new multi-“omics” techniques in immunology have revolutionized our molecular understanding of basic immune mechanisms. The extended and revisited concept of Inflamm-aging sheds a new light on the intricate relationships among immunosenescence and inflamm-aging. The new concept of inflamm-aging integrates different emerging experimental data such as those pertaining to the senescence-associated secretory phenotype and responses to damage. “Geroscience” is emphasising the role of immune changes during life in all manner of age-related chronic diseases such as cancer and neurodegenerative disorders. The field is poised to be in a position to translate these accumulated data into the clinical setting via a better understanding of the contribution of immunosenescence to age-associated pathologies, and their prevention by appropriate interventions. This new edition of the Handbook seeks to encompass the current state of our knowledge on the multitude of those changes to immunity related to aging, with contributions from experts in both basic research and clinical areas. Abstract

Abstract

Zur Untersuchung des Einflusses von tubulärem HIF auf die renale EPO-Produktion generierte Navid Farsijani ein genetisches Mausmodell, in dem durch tubuläre Vhl-Ablation, tubuläre Epithelzellen HIF überexprimieren. Die tubuläre Vhl-Ablation führte zu einer prompten Suppression der renalen EPO-Produktion. Anhand weitere konditioneller Mausmodelle zeigt der Autor, dass die Entwicklung der Anämie von HIF-abhängig ist, proximale Tubuluszellen für die Regulation von EPO im Nephron verantwortlich sind und die tubuläre Vhl-Ablation mit einer Reduktion der mitochondrialen Masse sowie einer Erhöhung des kortikalen Gewebssauerstoffpartialdruckes (PtO2) einhergeht. Dies spricht für eine multi-zelluläre Regulation der renalen EPO-Synthese, in der eine Homöostase zwischen tubulären Epithelzellen und REPC für eine adäquate EPO-Produktion der Niere notwendig ist. Zudem könnte der EPO-suppressive Effekt von tubulärem HIF einen pathophysiologischen Faktor in der Entstehung von renalen Anämien darstellen. Während sich Befürworter von Parallelimporten hohe Einsparungen für das Gesundheitswesen versprechen, befürchten die Gegner, dass die Einsparungen für den Patienten gering, die Auswirkungen auf den Forschungsstandort Schweiz aber gravierend wären. C. Poget untersucht, wie sich Parallelimporte von Arzneimitteln in Skandinavien auf Arzneimittelpreise und -ausgaben auswirken, und ob die Rahmenbedingungen in der Schweiz einem wirksamen Wettbewerb unter Parallelimporteuren zuträglich sind.

Abstract

In this book we provide insights into liver – cancer and immunology. Experts in the field provide an overview over fundamental immunological questions in liver cancer and tumorimmunology, which form the base for immune based approaches in HCC, which gain increasing interest in the community due to first promising results obtained in early clinical trials. Hepatocellular carcinoma (HCC) is the third most common cause of cancer related death in the United States. Treatment options are limited. Viral hepatitis is one of the major risk factors for HCC, which represents a typical “inflammation-induced” cancer. Immune-based treatment approaches have revolutionized oncology in recent years. Various treatment strategies have received FDA approval including dendritic cell vaccination, for prostate cancer as well as immune checkpoint inhibition targeting the CTLA4 or the PD1/PDL1 axis in melanoma, lung, and kidney cancer. Additionally, cell based therapies (adoptive T cell therapy, CAR T cells and TCR transduced T cells) have demonstrated significant efficacy in patients with B cell malignancies and melanoma. Immune checkpoint inhibitors in particular have generated enormous excitement across the entire field of oncology, providing a significant benefit to a minority of patients.  .

Abstract

Human biology has now entered into a phase of post-genomics and it might not be an exaggeration to say that the major outcome of the human genome sequencing has finally been to open the way to the exploration of the proteome-proteomics. Proteins are the functional output of genes and there are two main expected outcomes from human proteomics. The first is to discover new molecular markers for early diagnosis and profiling of pathologies. The second is to decipher the intracellular signaling pathways leading to the initiation and progression of pathologies, for the identification of new targets and the development of innovative therapeutic strategies. This is clearly a promising challenge that this book explores through a series of ongoing experiences and projects representative of the new era in which biology and medicine have now entered.
